dc.description.abstract | Bacterial cyclic glucans are glucose polymers that concentrate within the periplasm of alpha-proteobacteria. These molecules are necessary to maintain the homeostasis of the cell envelope by contributing to the osmolarity of Gram negative bacteria. Here, we demonstrate that Brucella β 1,2 cyclic glucans are potent activators of human and mouse dendritic cells. Dendritic cells activation by Brucella β 1,2 cyclic glucans requires TLR4, MyD88 and TRIF, but not CD14. The Brucella cyclic glucans showed neither toxicity nor immunogenicity compared to LPS and triggered antigen-specific CD8+ T cell responses in vivo. These cyclic glucans also enhanced antigen-specific CD4+ and CD8+ T cell responses including cross-presentation by different human DC subsets. Brucella β 1,2 cyclic glucans increased the memory CD4+ T cell responses of blood mononuclear cells exposed to recombinant fusion proteins composed of anti-CD40 antibody and antigens from both hepatitis C virus and Mycobacterium tuberculosis. Thus cyclic glucans represent a new class of adjuvants, which might contribute to the development of effective antimicrobial therapies. | es_ES |
dc.description.abstract | Los glucanos cíclicos bacterianos son polímeros de glucosa que se concentran en el periplasma de las alfa-proteobacterias. Estas moléculas son necesarias para mantener la homeostasis de la envoltura celular contribuyendo a la osmolaridad de las bacterias Gramnegativas. Aquí demostramos que los glucanos cíclicos de Brucella β 1,2 son potentes activadores de las células dendríticas humanas y de ratones. La activación de las células dendríticas por los glucanos cíclicos de Brucella β 1,2 requiere TLR4, MyD88 y TRIF, pero no CD14. Los glucanos cíclicos de Brucella no mostraron ni toxicidad ni inmunogenicidad en comparación con el LPS y desencadenaron respuestas celulares CD8+T específicas de antígeno in vivo. Estos glucanos cíclicos también aumentaron las respuestas específicas de los antígenos de las células CD4+ y CD8+T, incluyendo la presentación cruzada de diferentes subconjuntos de CD humana. Los glucanos cíclicos de Brucella β 1,2 aumentaron la memoria de las respuestas de las células CD4+T de las células mononucleares de la sangre expuestas a proteínas de fusión recombinante compuestas de anticuerpos anti-CD40 y antígenos tanto del virus de la hepatitis C como del Mycobacterium tuberculosis.
